Take-home

A new phase 3 trial puts divarasib head-to-head against existing KRAS G12C inhibitors, with encouraging results.

In previously treated KRAS G12C-mutated advanced NSCLC, divarasib significantly improved median progression-free survival (9.8 vs 5.7 months; HR 0.58) and overall survival (17.7 vs 12.0 months; HR 0.56) compared with sotorasib or adagrasib.

Response rates were also higher (58.4% vs 32.3%), with fewer grade 3–4 treatment-related adverse events (24.6% vs 34.5%).
