Safety of combining radiation therapy and antibody-drug conjugates in patients with breast cancer: a systematic review and meta-analysis
This systematic review and meta-analysis of 29 studies involving 3,817 patients evaluated the safety of combining radiotherapy (RT) with antibody-drug conjugates (ADCs) in #BreastCancer Severe extracranial toxicities were uncommon with T-DM1, T-DXd, and sacituzumab govitecan (SG). However, intracranial RT revealed a consistent radiation necrosis safety signal with T-DM1, not observed with T-DXd or SG. Cribiform and intraductal carcinoma in prostate cancer-implications for risk stratification and therapeutic decision making (REVIEW)
Intraductal carcinoma of the prostate (IDC) and invasive cribriform carcinoma are aggressive histological patterns associated with adverse pathology, metastasis, and prostate cancer mortality. Frequently underdetected on biopsy, they exhibit PTEN loss, genomic instability, DNA-repair alterations, and immunologically cold tumour microenvironments. Although these features suggest potential biomarker-directed therapies, evidence remains insufficient. Standardized reporting, improved detection, and prospective studies are essential to guide risk stratification and treatment decisions. #ProstateCancer

Atezolizumab approved with fluoropyrimidine + oxaliplatin for adjuvant Stage III dMMR Colon Cancer (adults & peds ≥2 yrs).
Subcutaneous Tecentriq Hybreza also approved (≥12 yrs, ≥40 kg). Approval based on ATOMIC (NCT02912559): DFS HR 0.50 (95% CI 0.35–0.73; p=0.0001). 3-yr DFS ~86.3% vs 76.2%. Median DFS NR either arm.

Advances in Systemic Treatment of Breast Cancer (Review)
Since 2020, systemic #BreastCancer therapy has advanced through targeted treatments, antibody–drug conjugates, immunotherapy, and biomarker-guided strategies. CDK4/6 inhibitors remain central in hormone receptor–positive disease, while novel agents address endocrine resistance. Trastuzumab deruxtecan is reshaping HER2-positive treatment, and immunotherapy benefits selected triple-negative cases. Precision medicine increasingly supports risk-adapted treatment. Key challenges include optimizing sequencing, refining predictive biomarkers, integrating circulating tumor DNA, and ensuring equitable access to innovative, often costly therapies.

177Lu-Prostate-Specific Membrane Antigen Neoadjuvant to Stereotactic Ablative Radiotherapy for Oligorecurrent Prostate Cancer (LUNAR)
The phase II LUNAR trial investigated whether adding PSMA-targeted radioligand therapy (¹⁷⁷Lu-PNT2002) before stereotactic body radiotherapy (SBRT) improves outcomes in oligorecurrent hormone-sensitive prostate cancer. Among 92 randomized patients, combination therapy significantly prolonged median progression-free survival from 7.4 to 17.6 months.

Effect of Screening with Multicancer Early-Detection Test on Late-Stage Cancer Diagnosis
A large randomized English🇬🇧 trial involving 142,250 adults aged 50–77 evaluated annual multicancer early-detection blood testing alongside usual care. After three screening rounds, there was no significant reduction in stage III/IV cancers compared with usual care alone (incidence rate ratio 1.03; P=.63). Stage IV cancer incidence was lower with screening (IRR 0.86), although the result requires further follow-up. Trial-related adverse events were uncommon and nonserious

FDA NEWS: HER2CLIMB-05 Update
On October 7, 2026, the FDA approved tucatinib with trastuzumab and pertuzumab as maintenance treatment for adults with unresectable locally advanced or metastatic HER2-positive breast cancer after induction therapy. In HER2CLIMB-05, tucatinib significantly improved median PFS versus placebo (24.9 vs 16.3 months; HR 0.64, p<0.0001) in 654 patients, including those with brain metastases. OS data were immature.
